IL-21/IL-21R Promotes the Pro-Inflammatory Effects of Macrophages during C. muridarum Respiratory Infection

چکیده
Interleukin-21 and its receptors (IL-21/IL-21R) aggravate chlamydial lung infection, while macrophages (Mφ) are one of the main cells infected by chlamydia source inflammatory cytokines. Therefore, it is particularly important to study whether IL-21/IL-21R aggravates respiratory infection regulating Mφ. Combined with bioinformatics analysis, we established an IL-21R-deficient (IL-21R−/−) mouse model Chlamydia muridarum (C. muridarum) tract in vivo, studied C. muridarum-stimulated RAW264.7 addition rmIL-21 vitro, conducted adoptive transfer experiments clarify association between IL-21R−/− mice showed lower infiltration pulmonary total Mφ, alveolar macrophages, interstitial compared WT following infection. Transcriptomic analysis suggested that M1-related genes downregulated IL-21R deficiency affects Mφ-mediated response during In vivo verified mice, M1-type CD80+, CD86+, MHC II+, TNFα+, iNOS+ Mφ decreased, there were no differences M2-type CD206+, TGF-β+, IL-10+ ARG1+ administration promoted levels iNOS-NO expression IL-12p40 TNFα, but had effect on TGFβ or IL-10. Further, M1-like bone marrow-derived derived from unlike those effectively protected recipients against induced relieved pathology. These findings help understanding mechanism which exacerbates promoting proinflammatory

The Effect of MS14 on Production of Pro-inflammatory Cytokines by Macrophages
Objective(s) Using herbal medicines as a complementary treatment method is increasing in wide variety of diseases. MS14-an herbal-marine preparation-is reported to have anti-inflammatory and immunomodulatory activities; however, the mechanism underlying its therapeutic effect is not known. Macrophages play an important role in host defense mechanisms and carry out their role by producing vario...
متن کاملIL-21/IL-21R in Autoimmune Diseases
Interleukin-21 (IL-21), the member of the γ-chain-related cytokine family, and its receptor (IL-21R), has been evidenced involving in the immune response, resulting in the development of autoimmune diseases, infectious diseases and tumors. This short communication reviews an overall outline of its genetic variants and expression in autoimmune diseases. It focuses on a series of recent studies t...
متن کاملDC-Derived IL-10 Modulates Pro-inflammatory Cytokine Production and Promotes Induction of CD4+IL-10+ Regulatory T Cells during Plasmodium yoelii Infection
The cytokine IL-10 plays a crucial role during malaria infection by counteracting the pro-inflammatory immune response. We and others demonstrated that Plasmodium yoelii infection results in enhanced IL-10 production in CD4+ T cells accompanied by the induction of an immunosuppressive phenotype. However, it is unclear whether this is a direct effect caused by the parasite or an indirect consequ...
متن کاملDiacerein inhibits the pro-atherogenic & pro-inflammatory effects of IL-1 on human keratinocytes & endothelial cells
We investigated IL-1-induced regulation of genes related to inflammation and atherogenesis in human keratinocytes and endothelial cells, and if 'diacerein', an oral IL-1 inhibiting drug currently approved for use in osteoarthritis, would reverse IL-1's effects on these cells. Primary human keratinocytes and coronary artery endothelial cells were treated with either IL-1α or IL-1β, with and with...
